Just got the bike back from the dealer for a new front sprocket. I notice that the left side wheel adjustment is in between the second and third marks, but the right is ON the third mark. Is this a big deal? Are these marks trustworthy for setting alignment? Thanks!

Nice little tool.... the same thing can be accomplished with a yard stick or piece of metal stock however. Just lay it flat against the rear sprocket (not on chain or sprocket bolts) and align it with the front one.
The laser can be helpful if you buy one of those tools, but again it's not necessary. Your local hardware store will carry yard sticks for around $3 a piece. Assuming you can use one hand to hold the stick and one hand to crank the adjustment bolt it's one of the easiest and most accurate ways to align things. And for what it's worth; the little marks on the swingarm are close, but not totally accurate. The difference may be negligible to some people and it may be an issue for others, for me it's usually close enough.

Found the laser tool. Too rich for my blood. I'll stick with the simple rod tool. Thanks for the input guys.
